Osteoarthritis is a chronic degenerative disease that occurs due to an imbalance between the production and decomposition of cartilage and reduces the quality of life by accompanied by pain and swelling. Drugs used to treat osteoarthritis have side effects when taken for a long period of time, so the demand for health functional foods made from natural products that are safe and can be taken for a long time is increasing. Therefore, this study evaluated the efficacy of low-molecular-weight collagen (valine-glycine-proline-hydroxyproline-glycine-proline-al-anine-glycine; LMC) extract for osteoarthritis. This was confirmed through chondrocytes treated with H2O2 or LPS and animal models injected with monosodium iodoacetate (MIA) to induce degenerative tubular inflammation. Research results showed that LMC showed a protective effect by reducing cartilage degradation in both chondrocytes and animal models. This increases the levels of anabolic factors in cartilage tissue, including Aggrecan, Collagen type I, Collagen type II, TIMP-1 and TIMP-3, and p-smad3, while reducing catabolic factors such as of MMP-3 and MMP-13. Additionally, LMC effectively inhibited the activation of inflammatory and apoptotic pathways in both chondrocytes and animal models. This reduced inflammatory cytokines (TNF-α, IL-1β, IL-6), PGE2, NO, and reduced the expression of apoptosis (JNK, c-Fos, c-Jun, FADD, Caspase 8, Bax, Caspase 3) and inflammation (IκBα, COX-2, p65) factors. In addition, the efficacy of low-molecular-weight collagen was confirmed through H&E, Micro-CT, and morphological observation in an animal model in which degenerative arthritis was induced, and the results showed that the bone structure of animals that consumed low-molecular-weight collagen extract was dense and cartilage cells were densely arranged. Based on these results, it was confirmed that LMC has potential as a natural material to improve osteoarthritis.
