    Chapter 3

Non-alcoholic fatty liver disease (NALFD) is lipid accumulation in liver without alcohol, it affects many people worldwide. NALFD occur to be associated with metabolic syndrome disease such as obesity, insulin resistance, hyperlipidemia, and diabetes. However, there is no pharmacologic therapies for NALFD. Recently, there is increasing reports that several natural plant can inhibit lipid accumulation in hepatocytes. Bay laurel
(Laurus nobilis L.) leaves have been used in traditional medicine for rheumatism, stomachache, emetic, skin
rashes, and earaches. Our objective was to investigate effect of bay laurel leaves water extract (BLW) on high fructose, high fat (HFHF) diet in a mouse model of NAFLD. Mice were ranomly divided into six groups: a normal AIN93G diet group (normal control;NC), a 10% high fructose+60% high fat diet group (HFHF), a 10% high fructose+60% high fat diet including 30 mg/kg body weight of silymarin extracts (positive control;HFHF+PC), a 10% high fructose+60% high fat diet including 50 mg/kg body weight of BLW group (HFHF+50), a 10% high fructose+60% high fat diet including 100 mg/kg body weight of BLW group (HFHF+100), a 10% high fructose+60% high fat diet including 200 mg/kg body weight of BLW group (HFHF+200). The mice were fed high fat diet add each concentration of BLW which is based on AIN93G and were fed gavage of 10% fructose solution. After 12 weeks, mice were sacrificed. Serum ALT, AST, and GGT levels in BLW supplementation group were significantly decreased compared with the HFHF group. Hepatic TC, TG, SOD, CAT, and GPx levels were significantly decreased in the HFHF+100 and HFHF+200 groups compared to the HFHF group. Expression of proteins related to hepatic lipogenesis was decreased in the BLW supplementation groups compared to the HFHF group. Furthermore, the expression of proteins related to hepatic inflammation and apoptosis was reduced in the BLW supplementation groups compared to the HFHF group. These results suggest that BLW could potentially useful in the treatment of NAFLD due to its inhibitory effects on hepatic lipogenesis, hepatic inflammation, and hepatic apoptosis.

    Chapter 2

Non-alcoholic fatty liver disease (NAFLD) is the accumulation of more than 5% fat in the liver without alcohol and a hepatic lipid metabolism disorder cause by various cause in worldwide. Free fatty acid (FFA) could be taken up by hepatocytes and provide as important source for triglyceride (TG) synthesis in hepatocytes. NAFLD is no approved pharmacologic therapies, so treatment for NAFLD has focused on lifestyle change. Many researches were reported that natural plants can protect fatty liver disease. Bay laurel (Laurus nobilis L.) contain various phytochemical agents and have been used in tradition medicine as an anti-emetic, and to treat earaches, stomach aches, rheumatism, and skin rashes. However, it is unclear if bay laurel leaves water extract (BLW) was able to protect against NAFLD in vitro. We propose that whether bay laurel leaves water extract (BLW) could be prevent NAFLD. HepG2 cells were treated with BLW (50, 100, and 200 μg/mL) and isoquercetin (IQ; 10, 20 mM) for 4 h, after free fatty acid solution (palmitic acid : oleic acid, 1:2) was treated for 24 h. We were determined by the lipid accumulation using oil red O staining, antioxidant activities, MDA levels, and protein expression using western blot assay. Lipid accumulation in HepG2 cells treated with FFA were increased in the control group, whereas BLW and IQ pre-treatment were reduced lipid accumulation to a level comparable to that in the control group. Cellular SOD, CAT, GPx, and MDA levels were similar in the BLW and IQ pre-treatment groups to those in the control group. Expression of CD36, SREBP-1c, and FASN as assessed by western blot was decreased in the BLW and IQ pre-treatment groups, while the expression of p-AMPK/AMPK, p-ACL/ACL, p-ACC/ACC, PPAR-α, and CPT1A was increased in the BLW and IQ pre-treatment groups. Furthermore, the expression of proteins of p-IκB/IκB, p-NF-κB/NF-κB, COX-2, and iNOS was reduced in BLW and IQ pre-treatment groups compared to the control group. In addition, the expression of proteins of p-JNK/JNK, BAX/Bcl-2, CD95, FADD, C-CAS8, and C-CAS3 was reduced in BLW and IQ pre-treatment groups compared to the control group. These results suggest that BLW and IQ protect against FFA induced fatty liver disease by attenuating lipid accumulation, inflammation, and apoptosis in hepatocytes.
