      Objective: This study evaluated the antibacterial and antibiofilm activities of the wasabiderived compound 6-(methylsulfinyl)hexyl isothiocyanate (6-MITC) against Streptococcus mutans. Methods: The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of 6-MITC against S. mutans were determined. Antibiofilm activity of 6-MITC was confirmed by human saliva-coated biofilm formation assay. Plates were incubated for 48 h in 1% sucrose-containing saliva medium with bacteria and 6-MITC. After staining with 0.1% crystal violet, the biofilms were dissolved in 20% acetic acid, and absorbance was measured. Results: The MIC and MBC of 6-MITC against S. mutans were 0.125 mg/mL and 0.5 mg/mL, respectively. Biofilm formation was reduced in a concentration-dependent manner by 6-MITC, and, with statistically significant inhibition observed at concentrations ≥0.25 mg/mL (p<0.05). Conclusions: Wasabi-derived 6-MITC exhibits antibacterial and antibiofilm activities against S. mutans. These findings suggest that 6-MITC has potential as a natural candidate for the development of oral hygiene products, antibacterial agents, and infection-control formulations.
